Biological insect control using Metarhizium anisopliae: morphological, molecular, and ecological aspects Ciência Rural
Tiago,Patricia Vieira; Oliveira,Neiva Tinti de; Lima,Elza Áurea de Luna Alves.
Microbial control of insects is based on the rational use of pathogens to maintain environmentally balanced pest population levels, and Metarhizium anisopliae has been the most studied and most utilized fungal species for that purpose. The natural genetic variability of entomopathogenic fungi is considered one of the principal advantages of microbial insect control. The inter- and intraspecific variability and the genetic diversity and population structures of Metarhizium and other entomopathogenic fungi have been examined using ITS-RFLP, ISSR, and ISSP molecular markers. The persistence of M. anisopliae in the soil and its possible effects on the structures of resident microbial communities must be considered when selecting isolates for biological insect...

Bovine μ-calpain (CAPN1) gene polymorphisms in Brangus and Brahman bulls JBAG
Soria,LA; Corva,PM; Huguet,MJ; Miño,S; Miquel,MC.
The bovine CAPN1 gene encodes the large subunit of μ-calpain, which is thought to be one of the most important enzymes involved in postmortem beef tenderization. Three SNPs in CAPN1 (SNP 316, SNP 530 and SNP 4751) have been associated with beef tenderness in different beef cattle breeds. The objective of this work was to implement genotyping strategies for CAPN1 markers as part of a project pursuing the identifi cation and validation of molecular markers associated with bovine meat quality and composition. Three PCR-RFLP methods were designed to determine genotypes of 64 bulls (11 Angus, 43 Brangus and 10 Brahman). Unexpected patterns resulting from the PCR-RFLP analysis at SNP 316 and SNP 530 were resolved by cloning and sequencing and lead to...

Challenges and prospects of population genetic studies in terns (Charadriiformes, Aves) Genet. Mol. Biol.
Faria,Patrícia J.; Baus,Erika; Morgante,João S.; Bruford,Michael W..
Little information is available about the population structure of communally nesting terns (Sternidae) and skimmers (Rynchopidae) throughout the world. In order to fill this gap, a survey of molecular markers was carried out for six species of terns (Anous stolidus, Sterna hirundinacea, S. fuscata, S. superciliaris, Thalasseus maximus and Phaetusa simplex) and one species of skimmer (Rynchops niger). First, we describe the results of the construction of genomic DNA libraries and document problems encountered during this procedure. Secondly, we tested the cross-amplification of 18 microsatellite loci previously described for related species (the number of polymorphic loci ranged from three to seven). Thirdly, we tested the usefulness of mtDNA (control...

Bertini,Cândida H.C. de Magalhães; Schuster,Ivan; Sediyama,Tocio; Barros,Everaldo Gonçalves de; Moreira,Maurílio Alves.
Genetic diversity and the relationship between varieties are of great importance for cotton breeding. Our work was designed to estimate the informativeness of the cotton (Gossypium hirsutum L.) simple sequence repeat (SSR) microsatellite locus and to estimate the genetic distance between 53 cotton cultivars as well as to select a set of SSR primers able to differentiate between the 53 cotton cultivars studied. After extracting DNA from the 53 cultivars and characterized it using 31 pairs of SSR primers we obtained a total of 66 alleles with an average of 2.13 alleles per SSR locus and values of polymorphism information content (PIC) varying from 0.18 to 0.62, the dissimilarity coefficient varying from zero to 0.41. Statistical analysis using the unweighted...

Characterization of Brazilian soybean cultivars using microsatellite markers Genet. Mol. Biol.
Priolli,Regina Helena Geribello; Mendes-Junior,Celso Teixeira; Arantes,Neylson Eustáquio; Contel,Eucleia Primo Betioli.
Microsatellite markers or SSR (Simple Sequence Repeats) have proved to be an excellent tool for cultivar identification, pedigree analysis and the evaluation of genetic distance among organisms. Soybean cultivars have been characterized mainly by morphological and biochemical traits. However, these traits have not been sufficient to characterize the large number of cultivars eligible to receive protection under the Brazilian Cultivar Protection Act. In order to define new soybean cultivar markers, the alleles of twelve SSR loci of 186 Brazilian soybean cultivars were studied by estimating the variation in their size range and their respective frequencies. On average, 5.3 alleles per locus were detected, with a mean genetic diversity of 0.64 ± 0.12. These...

Silva,Ana Veruska Cruz da; Amorim,Julie Anne Espíndola; Vitória,Marina Ferreira da; Ledo,Ana da Silva; Rabbani,Allivia Rouse Carregosa.
ABSTRACT The state of Sergipe is the largest mangaba producer, which is a fruit native to Brazil, and has cultural, social and economic importance in its area of occurrence. It is an endangered species due to human actions, and despite its economic potential, there are still no commercial plantations. The study was carried out in order to characterize trees, fruits and the genetic diversity of natural populations of mangaba in Sergipe, Brazil. Fruits from Abaís Beach/Estância (AB) presented, on average, twice the vitamin C content (414.81 mg of vit. C/100g), when compared with the others. The use of ISSR primers was efficient in estimating the genetic similarity of populations. The primers clustered the populations of mangaba according to their origin,...

Chromosomal localization of selected SCARs converted from new RAPD, ISSR and R-ISSR markers linked to rye (Secale cereale L.) tolerance to nutrient deprivation stress identified using bulk segregant analysis Electron. J. Biotechnol.
Smolik,Milosz.
Background: An adaptive mechanism in plant roots is initiated in the event of nitrogen and potassium deficiency, and it facilitates the active uptake of these elements in order to ensure plant growth and survival in stress conditions. Signaling and transduction of signals in response to changing nitrogen and potassium concentrations is a complex process, affected by interactions between various gene expression products, and often subjected to modifications. Results: In order to identify genotypic differences between phenotypes of two populations of recombinant inbred rye lines (153/79-1 x Ot1-3 and Ot0-6 x Ot1-3) in response to nutrition stress caused by nitrogen and potassium deficiency at the seedling stage, bulk segregant analysis was utilized....

Comparative analysis of genetic diversity among the maize inbred lines (Zea mays L.) obtained by RAPD and SSR markers BABT
Souza,Silvia Graciele Hülse de; Carpentieri-Pípolo,Valéria; Ruas,Claudete de Fátima; Carvalho,Valdemar de Paula; Ruas,Paulo Maurício; Gerage,Antônio Carlos.
The RAPD and SSR markers were used to compare the genetic diversity among the 16 maize inbred lines. Twenty-two primers were used in the RAPD reactions, resulting in the amplification of 265 fragments, while 16 pairs of SSR primers resulted in 75 fragments. The similarity based on Dice coefficient for the RAPD ranged from 53 to 84% and for the SSR from 11 to 82%. The dendrogram obtained by the RAPD showed five groups, while dendrogram obtained by the SSR showed three groups and one isolated line. The association constructed from the markers and the principal coordinate’s analysis separated lines into two groups according to endosperm color, either orange or yellow. The RAPD were effective to validate pedigree data, while the SSR were effective to recognize...

Comparative linkage mapping of Oryza glumaepatula and Oryza sativa interspecific crosses based on microsatellite and expressed sequence tag markers Genet. Mol. Biol.
Rangel,Priscila Nascimento; Brondani,Rosana Pereira Vianello; Coelho,Alexandre Siqueira Guedes; Rangel,Paulo Hideo Nakano; Brondani,Claudio.
Molecular linkage maps representing the rice genome have been an important tool for breeding programs because they allow the elucidation of polygenic traits and are an efficient tool for monitoring wild introgressions in interspecific crosses. Common markers among rice genetic maps are important in defining the homology of chromosomes and the synteny between genomic target regions. We used 148 markers (expressed sequence tags, microsatellites and single nucleotide polymorphisms) to construct a molecular linkage map based on co-dominant markers for an interspecific backcross population using a wild rice (Oryza glumaepatula) from Brazil and performed a comparative analysis with other interspecific maps. The comparative analysis revealed a Spearman...

Studies on macromolecules isolated from marine algae suggested sulfated polysaccharides (SPs) as possible molecular markers for species. We evaluated isolated and fractionated SPs from the green marine algae Caulerpa cupressoides, C. prolifera and C. racemosa collected at Pacheco Beach, as possible taxonomic molecular indicators. Total SPs were extracted with papain in 100 mM sodium acetate buffer (pH 5.0) containing cysteine and EDTA (both 5 mM), followed by ion-exchange chromatography on DEAE-cellulose using a NaCl gradient. The obtained fractions were analyzed by 0.5% agarose gel electrophoresis. Anticoagulant assays employing normal human plasma and standard heparin (193 IU mg-1) by the activated partial thromboplastin time (APTT) test were also...

Comparison of PCR-based molecular markers for the characterization of Proteus mirabilis clinical isolates BJID
Michelim,Lessandra; Muller,Gabriela; Zacaria,Jucimar; Delamare,Ana Paula Longaray; Costa,Sérgio Olavo Pinto da; Echeverrigaray,Sergio.
Proteus mirabilis is one of the most important pathogens associated with complicated urinary tract infections (acute pyelonephritis, bladder infections, kidney stones) and bacteremia, affecting patients with anatomical abnormalities, immunodeficiency, and long-term urinary catheterization. For epidemiological purposes, various molecular typing methods, such as pulse-field gel electrophoresis (PFGE) or ribotyping, have been developed for this pathogen. However, these methods are labor intensive and time-consuming. We evaluated the discriminatory power of several PCR-based fingerprinting methods (RAPD, ISSR, ERIC-PCR, BOX-PCR and rep-PCR) for P. mirabilis clinical isolates. Typing patterns and clustering analysis indicated that RAPD, BOX-PCR and ERIC-PCR...

Comparison of RAPD, RFLP, AFLP and SSR markers for diversity studies in tropical maize inbred lines Genet. Mol. Biol.
Garcia,Antonio A. F.; Benchimol,Luciana L.; Barbosa,Antônia M. M.; Geraldi,Isaias O.; Souza Jr.,Cláudio L.; Souza,Anete P. de.
In order to compare their relative efficiencies as markers and to find the most suitable marker for maize diversity studies we evaluated 18 inbred tropical maize lines using a number of different loci as markers. The loci used were: 774 amplified fragment length polymorphisms (AFLPs); 262 random amplified polymorphic DNAs (RAPDs); 185 restriction fragment length polymorphisms (RFLPs); and 68 simple sequence repeats (SSR). For estimating genetic distance the AFLP and RFLP markers gave the most correlated results, with a correlation coefficient of r = 0.87. Bootstrap analysis were used to evaluate the number of loci for the markers and the coefficients of variation (CV) revealed a skewed distribution. The dominant markers (AFLP and RAPD) had small CV values...
